Abstract

INFLUENCE OF PULSE ELECTROMAGNETIC FIELD ON THE VIABILITY OF ESCHERICHIA COLI in WATER model solutions

An important indicator of microbiological safety and quality, and production sanitary conditions are bacteria of Escherichia coli.

At excess of acceptable titer these microorganisms can cause to damage the finished product and even food poisoning.

The actual problem is the replacement of chemical methods of cleaning the water on physical nonchemical methods. This issue can be solved through the use of electric fields. It should be noted that the PEF technology is regarded as one of the most promising nonthermal methods to inhibit microorganisms in food and water while preserving color, flavor, structure and nutritional value.

The aim was to study the impact of actions pulse electromagnetic fields on enterobacteria, such as Escherichia coli culture solution in modeling water.

Methods. In Kharkiv Polytechnic Institute "NTU KPI" at the Department of Engineering Electrophysics were conducted experimental studies on the cultivation model solutions Escherichia coli in the water. The studies were conducted using experimental installation developed of National Technical University «Kharkiv Polytechnic Institute».

Sterilization water in an autoclave at t = 1190C for two hours. In sterelizovanu water made the required number of E. coli bacteria to get 106 and 108 breeding КОУ / sm3. Then the sterile working chamber volume 150sm3 Closed-made solutions modelli.

Model solutions of whey treated at voltage of 15 ... 30 kW/cm3, treatment time 10 ... 20 s.

After processing electromagnetic modeling solutions in microbiological laboratories happened quantitative calculation of surviving bacteria by direct counting the colonies on dense nutrient medium (GOST 26670-91).

For the differentiation of Escherichia coli was used Endo medium where the bacteria growth appears in the form of red colonies with metal shine.

Results and Discussion. It was determined that PEF treatments with voltage of 10 kW/cm3 during  20 s is the most effective, because there was complete inactivation of Escherichia coli cells inoculated to water. Research results of milk serum model solution confirmed by public utility "SANEPIDSERVISOM" Kharkiv.

It was proved the possibility of heat treatment of water by non-thermal effects generated by the pulse action of electrical fields.

It was showed the perspective of using local PEF equipment at primary treatment of milk serum for microbial inactivation.
